An Electron Microscopic Study on the Effect of Isoniazid on the Mouse Sciatic Nerve

Abstract
|
|
|
In an attempt to elucidate the effect of isoniazid on the sciatic nerve of mouse, electron microscopic study was performed to examine the fine structure of sciatic nerve of mouse after administration of isoniazid. Mice were received daily 100mg and 200mg of isoniazid per kg body weight through gastric intubation and sacrified 2 and 4 weeks respectively.
The results were summerized as follows :
1. Morphological changes of isoniazid neuropathy were similar to those of Wallerian degeneration.
2. Myelin sheath of sciatic nerve affected by isoniazid was collapsed abnormally compared with control group.
3. The space formed by shrinking of axoplasm was observed between the axon and myelin sheath and also some vacuoles were shown in the axoplasm.
4. In the experimental group, the number of cell organelles in Schwann cell were slightly decreased than those of the control group, and some vacuoles were also appeared in the cytoplasm of Schwann cell. From these results, isoniazid induces peripheral neuropathy, especially in axon, myelin sheath and Schwann cells.
